Action: Inhibits leukotriene (LTD4) formation; leukotrienes exert their effects by increasing neutrophil, eosinophil migration; aggregation of neutrophils, monocytes; smooth muscle contraction, capillary permeability; these actions further lead to bronchoconstriction, inflammation, edema
Uses: Chronic asthma
Dosage and routes:
Asthma
  • Adult and child greater than or equal to 15 yr: PO 10 mg qd pm
  • Child 6-14 yr: PO 5 mg chew tab qd pm
Available Forms: Tabs 10 mg; tabs, chew 5 mg
Side effects/adverse reactions:
CNS: Dizziness, fatigue, headache
GI: Abdominal pain, dyspepsia, LFTs abnormalities
INTEG: Rash
MS: Asthenia
RESP: Influenza, cough, nasal congestion
Contraindications: Hypersensitivity
Precautions: Acute attacks of asthma, alcohol consumption, pregnancy (B), lactation, child <6 yr, aspirin sensitivity
Pharmacokinetics: Rapidly absorbed, peak 3-4 hr, half-life 2.7-5.5 hr; protein binding 99%; metabolized by liver, excreted via bile
Interactions:
  • Decreased montelukast levels: phenobarbital, rifampin
